Protein Isolation and BCA

We focused on isolating and quantifying protein samples. Proteins can be extracted from tissues or cultured cells by disrupting the cell barriers to release their contents in a process known as cell lysis. The method of protein extraction varies depending on the sample type, the localization of the target protein, the desired yield, and the intended downstream applications.

Western Blot

Western blot is a widely used analytical technique for detecting specific proteins in a sample. The process involves separating proteins by gel electrophoresis, transferring them onto a membrane, and probing the membrane with antibodies specific to the target protein. This allows for the visualization and quantification of the protein, providing valuable insights into its expression levels and molecular weight.

PCR

PCR (Polymerase Chain Reaction) is a powerful technique used to amplify specific DNA sequences, enabling detailed genetic analysis. In our experiment, we utilized PCR to examine the PER3 gene, which plays a role in regulating circadian rhythms. By analyzing variations in this gene, we could determine whether we were genetically predisposed to be night owls or early birds.
